Turn on Air Beam and start recording to look for the particular matter.
Go to first location
Identify tree
- Honey locust and Sugar Maples because they are the most common ones on the areas that i’ll be collecting data on.
Take the circumference in centimeters using your measuring tape
Pick out 1-2 leaves and put in ziplock bag and also write down on a piece of scratch paper your field number and put in ziplock bag with the leaf.
Take epicollect data and fill out the data
Move to next location
Repeat steps 2-5

Take out the first leaf you collected
Cover a small area about 1cm x 2cm on the back of the leaf with clear nail polish for the sugar maple leaf and for the honey locust leaf cover the whole leaf with nail polish since it is a smaller leaf.
Blow or shake leaf to dry it up and wait 30 secs or more until nail polish dries completely.
Put a piece of clear tape on top of the area covered with the nail polish without putting many fingerprints on the tape.
Press the tape down on leaf to get all the nail polish then remove tape.
Put the piece of tape on a clear slide while trying not to put fingerprints onto the tape and label slide with sharpie and put slide under a microscope.
On microscope change the focus to 400x take picture of first area so it’s easier to count the stomata.
Record on data sheet and move on to next area on the slide.
Repeat for the last area of the slide. For a total of 3 areas
Get the average count of stomata per area for the one leaf.
Repeat steps 1-10 for the rest of the leaves collected.
